I don't see WPMUDev's Defender/Defender Pro on the list of compatible security plugins. Has it been tested or are there any known incompatibilities?

We've tested the plugin Defender Security – Malware Scanner, Login Security & Firewall which is available on WordPress directory and works well with HMWP Ghost.
We have users who are using the PRO version of WPMUDev's Defender/Defender and didn't have any compatibility issue.
The plugins features are complementary and they will definitely boost your websites security.

I was just about to purchase but was made aware that there are some conflicts with WPMUDev hosting that break functionality on sites on their hosting to the point where they are no longer willing to edit Nginx rules for your plugin. Is this on your radar / do you have any plans on making the plugins compatible with WPMUDev hosting? That's where I host many client sites and would love to use your product.

I've been told by WPMUDev specifically that your plugin is not compatible with their hosting. I'm happy to share the screenshots of that conversation with you if you'd like.
