So the Tier 4 plan which allows M365, it says 25 signatures, is that 25 MS365 users of the company or each user gets 25 signatures even if we have 40 email accounts? Its not clarified anywhere.

Tier 4 allows you to create up to 25 signatures under your Newoldstamp account. Usually, 1 signature is created for 1 user. Also, if you have more users in your Entra ID, but you need signatures to be created just for 25 of them, then Tier 4 is a great option.
