Suppose I am creating a program with 100+ lessons which require perhaps 500 screens. Can I create one AI-generated course and then use the AI assistant inside that same project to generate additional chapters and lessons without consuming another monthly course generation? Or does each new AI-generated chapter count as another course generation credit?

Thanks for your question, the maximum length of content produced by AI is about 30 chapters. To do this, set your course length to "In depth". You then have the option to add additional chapters manually and these manual chapters do not take up any of your monthly quota, as they do not use any Ai engine.
It also depends on your definition of a "lesson". If you are looking for 100+ individual SCORM files, this will be 100+ individual credits, which can be purchased within the software to increase your monthly limit. If multiple lessons are combined to fit into one SCORM package/generated course, this will of course take up less credits.
